The supermarket giant has applied for planning permission to demolish 86-88 in The Parade - the former police station - and convert into a retail area.
They also plan to build two two-bedroom flats and two one-bedroom flats above it.
A decision is expected by August 11.
Previous plans to demolish the site and build a Tesco Express store were rejected by Wycombe District Council in 2009.
Jim Penfold, chairman of Bourne End Residents Association, said he saw few changes in the new application.
"I would urge the people of Bourne End to resist it as strongly as possible," he added.
